Ⅰ. 細菌のイオウ代謝と抗酸化防御機構の解明

βラクタム抗菌剤は、細菌の細胞壁合成を阻害する抗菌剤で、その優れた有効性や副作用の少なさから、多くの細菌感染症の治療に用いられています。なかでもカルバペネムと呼ばれるβラクタム抗菌剤は、従来のβラクタム抗菌剤が効かなくなった耐性菌に対する「最後の砦」とされています。これまでの研究から、細菌の硫黄代謝過程で生じる硫化水素が、細菌の薬剤耐性に寄与することが報告されていましたがその詳細なメカニズムは分かっていませんでした。今回、我々は硫化水素が超硫黄と呼ばれる分子に変換されると、カルバペネムを含むβラクタム抗菌剤を強力に分解・不活性化することを発見しました。構造解析の結果、分解された薬剤は、βラクタム環が開環し硫黄が付加した、カルボチオ酸という新規化合物であることが分かりました。このカルボチオ酸に対する高感度分析法を開発し、その生成動態を解析した結果、βラクタム抗菌剤は、細菌に取り込まれた後、菌体内の超硫黄分子によりカルボチオ酸へと分解され、さらにそれが菌体外に排出されていることを発見しました。今回の結果は、これまでに知られていない、細菌によるβラクタム抗菌剤の分解経路を明らかにした画期的な成果です。今後、菌体外に排出されたカルボチオ酸を指標(バイオマーカー)とすることで、細菌の超硫黄分子産生を阻害する化合物のスクリーニングを計画しています。このような化合物は、超硫黄分子に依存したβラクタム抗菌剤の自然耐性を弱める効果(アジュバント効果)を有することが考えられ、その結果、より低濃度のβラクタム抗菌剤での治療が可能になり、新たな耐性菌の出現が抑えられると期待されます。

I. Elucidation of Sulfur Metabolism and Antioxidant Defense Mechanism in Bacteria

Beta-lactam antibacterial agents are antibacterial agents that inhibit bacterial cell wall synthesis, and are used to treat many bacterial infections due to their excellent efficacy and minimal side effects. Among them, beta-lactam antibacterial agents called carbapenems are considered to be the "last resort" against resistant bacteria that have become ineffective against conventional beta-lactam antibacterial agents. It has been reported that hydrogen sulfide, which is produced in the process of sulfur metabolism in bacteria, contributes to bacterial drug resistance, but the detailed mechanism has not been elucidated. In this study, we found that hydrogen sulfide, when converted to a molecule called super-sulfur, strongly degrades and inactivates β-lactam antibacterial agents including carbapenems. Structural analysis revealed that the degraded drug was a new compound called carbothioic acid, in which the β-lactam ring is ring-opened and sulfur is added. As a result of developing a highly sensitive analytical method for carbothioic acid and analyzing its formation kinetics, we found that the β-lactam antimicrobial agent is taken up by bacteria, degraded to carbothioic acid by super-sulfur molecules inside the bacteria, and then discharged out of the bacteria. This is a groundbreaking result that clarified the degradation pathway of β-lactam antimicrobial agents by bacteria, which has never been known before. In the future, we plan to screen for compounds that inhibit the production of supersulfur molecules by bacteria by using the carbothioic acid discharged from the bacteria as an indicator (biomarker). Such compounds may have the effect of weakening the natural resistance of β-lactam antimicrobial agents dependent on super-sulfur molecules (adjuvant effect), and as a result, it is expected that treatment with lower concentrations of β-lactam antimicrobial agents will be possible and the emergence of new resistant bacteria will be suppressed.
